Measuring CDOM with a Fiber Optic Liquid Waveguide Capillary Cell
One's imagination is the limit with practical uses of WPI's Liquid Wavelength Capillary Cells(LWCC), also referred to in the fiber optic spectroscopy community as a Long Pathlength Flow Cell. This fiber optic sampling accessory for absorbance measurements combines increased optical pathlengths with small sample volumes making them ideal for water analysis such as CDOM.
Two optical fibers couple to this sampling accessory one to send light to the sample cell and the second to send light to a spectrometer. The purpose of this article is to describe a new cleaning procedure for thoroughly cleaning WPI flowcells, including Liquid Waveguide Capillary Cells (LWCCs), UltraPath flowcells and optical cuvettes. The image (right) shows a 3000 series LWCC with a MiniStar pump and the LWCC injection system placed on top of it.
Preparation of Chemicals
All chemical reagents should be of at least ACS-Grade, preferably HPLC-Grade. This procedure involves the use of caustic and flammable reagents. Consult the manufacturer’s MSDS for necessary safety precautions.
Cleaning Solution #1:
0.5M Potassium Hydroxide in 100% Ethanol (e.g.: 7 g KOH in 250mL EtOH). After thoroughly mixing, filter the solution through a 20µm pore size filter.
Cleaning Solution #2:
100% Methanol, HPLC grade

What is REMS?
The REMS AutoSampler automates measurements of electrical resistance of transepithelial, transendothelial or Caco-2 cell membranes being grown to confluence on microporous filters of high throughput screening (HTS) 24- and 96-well microplates. It is a PC-controlled, tissue resistance measurement system that offers reproducibility, accuracy and flexibility for this kind of measurement. The ability of the REMS AutoSampler to precisely locate the electrode results in highly reproducible TEER measurements. TEER measurements are stored in the computer as the electrode moves from one well to the next. WPI offers a variety of precious metals and wire. The table below compares the specifications of the various wires. The following hints are offered to help you understand the part number codes and choose the wire you need:
- Micro coaxial cables (MAXxxxx) are ideal for microelectrode fabrication and construction of similar research tools. The dual shielding eliminates electrical interference caused by radio frequencies (RF), electrostatic and microphonics (e.g., bending and vibration). Available with single or dual (twin) conductors.
- Thermocouple wire is for temperature monitoring. TCK3050 has a chromel/alumel pair, for use in inert or oxidizing atmosphere at 1500-1800 °F.
- Teflon-coated stainless steel (type 304) wire (SSTxxxx) is available in 25-ft and 50-ft lengths. The Teflon coating is 4µm thick.
